For my first rotation, I developed a protocol for generating neuromuscular organoids (NMOs) from human pluripotent stem cell (hPSC)-derived neuromesodermal progenitors (NMPs). By using NMPs corresponding to the hindbrain and the high thoracic region, we were able to emulate distinct specialty regions to create specific NMO types. Our protocol produced organoids that showed increasing eccentricity as they matured and possessed expression changes that demonstrated maturation into neuronal and muscle areas.
